Module 6: Streak for Isolation

-First, draw a circle on a piece of paper, put in the guide pattern, and then
practice using a pencil and a piece of paper. Show the instructor.

-Do the procedure using a tube containing at least two different kinds of bacteria.
-The idea is to spread the bacteria on the plate in such a fashion that the mass of bacteria
are separated into single bacteria. The single bacterium then grow into individual colonies,
clones of the original bacterium. If this colony is used to innoculate a plate then the subsequent
growth would be a pure culture. To be sure of obtaining a pure culture, a second streak for
isolation would be done and a colony from this second streak would be used as the source
of the pure culture.
- Remember, this method assumes the bacteria will separate into individual baceria. It is difficult
to tell if the colony arose from two bacteria that were stuck together.
-It is always good practice to know the names of the bacteria with which you are working.

Day 2
A successful streak for isolation shows nicely separated individual colonies of bacteria. It does not matter
in what "zone" they are found, as long as there are individual colonies..
