Preliminary: Bunsen burner.
Practice with the striker.
- Be sure to be able to get sparks.
- Pull up while pulling in on the striker.
Turn on at the source, listen for hissing at the burner.
Two controls on the burner affect the quality and quantity of the flame.
-The knob on the bottom controls the quatity of gas. Turn it till hissing
is heard.
-The main tube is unscrewed until enough air enters a the bottom to
give a
strong blue flame.
Tips:
-Be careful when the burners are on, especially when reaching across
the bench..
-It is easier to hear than see the flame.
-Tie back hair
-Keep the burners in front of the taped area on the bench.
The flame is used to sterilize the loop. Sterilize the loop by moving
the flame
from the handle area to the tip. This is to prevent splatter of the
material on the tip
before it can be killed.
-Be careful with the loop after flaming as it will still be very hot,
even it is no longer is glowing red.
-Use the tip of the inner blue flame. It is the hottest part of the
flame.
Module 5: Aseptic Transfer
The main goal is to move bacteria from one area to another in such
a manner as to prevent contamination.
Briefly, transfer bacteria from a slant, or plate, or broth onto a
fresh slant, plate or broth.
-Flame the loop
-Hold the tubes in one hand and then unscrew and hold the caps in the
other hand.
-Flame the lip of the tube by quickly passing the tube thru the flame.
--Do not heat the lip too long. Just pass it 3-4 times thru the flame.
Day 2
Did the bacteria deliberately placed on the media grow, or did other
bacteria also grow?
-Was the broth cloudy? Could you tell how many types of bacteria were
growing?
-How many different kinds of colonies were noted on the nutrient agar
plates?
