This is part of the IMViC tests. Some bacteria are able to utilize tryptophan and produce indole. This test indicates the presence of indole by a red layer (see arrows) that forms on top after the addition of Kovacs reagent.  The Kovacs reagent should be layered on top. Do not mix.
